Monomi joins Monokuma, both separated by the replica door in the middle. "If I lock the door from this side, I can still freely open and close it." Monokuma explains, demonstrating. "However, Monomi won't be able to open the door, no matter how hard she tries."
Monomi gives the knob some good tugs but can't do anything. "He's right!"
"If Monomi locked the door from the other side, I wouldn't be able to open it. I'd have to use a bazooka or something to kill her. Got it so far?" Monokuma waited to see some nods from the classmates. "Great! Now, let's look at the first scenario again."
Monokuma locks the door from his side, preventing Monomi from being able to open it on hers. He then leaves the key in the keyhole, opens the door, and joins her side, closing the door behind him. He punches Monomi across the face and tries to open the door again, but it's locked. "Now let me show another scenario." Monokuma bows.
Monokuma locks the door from his side and takes the key out of the keyhole. He then opens the door, and gets to the other side with Monomi, closing the door behind him. He then opens it easily. "See? The door will only remain locked after being opened if the key is in the keyhole. And finally, I have one more scenario to show."
Monokuma is now the only one at the door. He puts the key in, locks it, and leaves it in the keyhole. He walks to the other side and closes the door. He tries to open it, but can't. Monomi then walks by the side with the key in it and yanks it out of the hole. Monokuma tries to open it again, but it's still locked. Then Monomi opens the door from her side (without putting the key in) and shuts it immediately after. Monokuma tries the door again, and it opens.
"Sorry if that was confusing, I can take any questions if they're needed. Essentially, the door can be opened while it's locked on one side and will remain locked as long as the key is still in the keyhole. If the key is removed from the keyhole, the door will be unlocked on both sides once it's opened from the side it was locked on."
In summary:
-Locking the door from side A will make it unable to be opened on side B
-Locking the door from side B will make it unable to be opened on side A
-Locking the door from side A and then opening and closing it from side A will have the door remain locked on side B as long as the key is in the keyhole
-Locking the door from side A and then removing the key will still have the door be locked on side B as long as the door isn't opened from side A

"Yeah, we were told by someone - I want to say Solomon or Matt? - that the door was actually unlocked from the hallway side, remember? We only thought we couldn't open it. If it was locked from the outside and the key removed that would make sense."
