I think Shiina will be the main villain of the game...Here's why:
Judging by the cover arts we've seen so far, we can only assume these two are going to be important characters throughout the game, not just in one case. They can't be mentors: they're not prosecutors, and we know Edgeworth's mentor is Manfred Von Karma. They can't be sidekicks, as Edgeworth already has Gumshoe / Faraday / perhaps even Ema. So they have to be villains or at least some kind of antagonists.
Now, Rou might very well be the "final boss", but that would be way too obvious. He looks too much like two other hotheaded murderers, for starters. Secondly, he's said to have a deep hatred towards prosecutors. Thirdly, he just looks evil. Shiina, on the other hand, seems to be more cautious, more calculating... she could be the one plotting behind the scenes. Besides, that would make for an interesting plot twist, as she appears to be working under Rou. Finally, she's the typical white haired pretty girl. For all those reasons, her being a villain seems rather likely.
That sounds like everything I've thought of Shiina, she will look like if she worked for him and they both will be the antagonists until the last case when you'll discover Shiina's super evil plan.
Then Rou will find out that his hatred against prosecutors was actually her superduper ultra evil plan and him and Edgeworth will share some beers.
But that leaves some other questions: what kind of past could Shiina have with Edgeworth? What is the evil plan's purpouse? Why did that affect Rou? and finally How can we know that both Shiryuu and Shiina aren't evil?
We can't possibly know.
